Rheumatoid arthritis is a common chronic inflammatory arthritis, which is characterized by articular synovitis and bone and cartilage destruction. Controlling tissue damage caused by inflammation and slowing bone destruction are of great clinical significance in RA treatment.
The overexpression of angiogenic protein 4 (ANGPTL4) has been implicated in the pathogenesis of RA. ANGPTL4 was originally thought to be a regulator of lipid metabolism and hydrolyzed into N-terminal (nANGPTL4) and C-terminal (cANGPTL4) fragments in vivo. CANGPTL4 is involved in several lipid-independent processes, including angiogenesis and inflammation.
In the study, the team first observed a significant increase in the expression of angiogenic protein 4 in serum and synovial tissue of rheumatoid arthritis patients. In collagen-induced arthritis and adjuvant-induced arthritis animal models, treatment with CANGPTL4 neutralizes antibodies significantly alleviates disease severity, inhibits inflammatory progression and bone destruction. Data from adjuvant-induced transcriptome and proteome of adjuvant-induced synovial tissue in animal models suggest that CANGPTL4 neutralizing antibody therapy inhibits fibroblast-like synovial cell migration and inflammation-induced osteoclastic production.
In addition, the team found that CANGPTL4 neutralizing antibodies blocked fibroblast-like synovial cell invasion and migration-induced osteoclast activation.
In summary, the study revealed that angiogenic protein 4 is a potential biomarker in the treatment of rheumatoid arthritis, providing scientific basis for exploring new ways to prevent and prevent rheumatoid arthritis joint bone destruction.
