Inspired by heartbeats pumping blood, the research team proposed a new strategy for mechanically assisted "biological 3D printing +".First, they combined 3D printing technology to build hollow fiber hydrogel scaffolds (HHSs) with mechanically responsive, large-scale complex structures, and then used the scaffolds' mechanically responsive properties to load cells quickly, evenly, accurately and friendly.Based on this strategy, the cell-loaded scaffold can effectively promote the repair and functional reconstruction of refractory bone defects.
This strategy effectively solves the difficult problem of balancing cell activity and scaffold mechanical stability in extrusion biological 3D printing process, which can ensure the accuracy of 3D printing technology and maintain the high survival rate of load cells.
In this study, the team successfully constructed a height-adjustable hollow hydrogel scaffold using coaxial needles and "one-step" 3D printing technology without support, which can recover quickly at 80% compression strain and maintain a complete structure after 10,000 compression cycles.
In addition, the stent has mechanical responsiveness, and the response behavior can be controlled by compressive strain and the number of cycles.HHS can achieve rapid, precise and partitioned loading of cells under mechanical stimulation.The number of HHS-loaded cells increased 13 times compared to static conditions.
The "biological 3D printing +" strategy proposed in the study breaks through the limitations of biological 3D printing that bio-ink cannot construct hydrogel scaffolds with both lossless cells and excellent mechanical properties.As a proof of concept, HHS of loaded cells showed enhanced regenerative ability in repairing segmental and osteoporosis bone defects in rats, which is expected to provide new approaches for tissue regeneration and cell therapy.
