Study reveals epigenetic memory of antiviral memory B cells
Recently, Hou Baidong's research team and Zhu Bing's research team published a research paper in the journal Advanced Science, which found that antiviral memory B cells (MemB) have both adaptive and natural immune memory characteristics.
With the recent research on the role of epigenetics in cell fate determination and intergenerational stable inheritance, people have begun to explore the contribution of epigenetics to natural immune memory formation.
In order to investigate the mechanism of antiviral MemB formation, Hou's research team used Qβ-derived virus-like particles (Qβ-VLP) as model antigens in mice.It induces long-lived hair growth center (GC) reactions, and Qβ-VLP-induced MemB can be stabilized in mice for more than a year and bind to antigens with high affinity, which is highly similar to those identified in humans.
In order to study the epigenetic information changes during the formation of these cells and their contribution to MemB cell function, Hou's team collaborated with Zhu Bing's team to evaluate the function of memory B cells by RNA-seq and ATAC-seq.The study found an increase in the proportion of genes whose MemB is enhanced by transcription in secondary reactions, and is associated with increased chromatin access in resting MemB, suggesting that epigenetic changes may initiate the expression of these genes.Qβ-VLP-induced secondary response MemB enriched more viral response-related genes than common protein antigen specificity, most of which were related to innate immune signaling.
Since natural immunity is an evolutionarily older form of immunity, adaptive immune systems retain some of the immune memory of natural immune cells after acquiring the ability to rearrange the V(D)J gene.This study sheds light on the epigenetic memory formation process in antiviral MemB, providing clues to the mechanisms of long-lasting immune memory formation.
