Recently, Zhang Jiwan team of Shanghai Institute of Pharmaceutical Research of Chinese Academy of Sciences and Wang Shujun team of Shenyang University of Pharmaceutical Sciences built a novel nano-grid structure carrier with cyclodextrin as the basic structure unit.The study was published in Microscale: Structure.
Based on the ordered structure of cyclodextrin metal organic skeleton, the research team cross-linked the borinated organic skeleton (BOF) with symmetric borate cross-linking agent according to the hydroxyl activity difference at different sites on cyclodextrin unit.Borate-bonded crosslinking agents give BOF a pH/H2O2 double responsiveness, that is, stable in physiological pH environment, rapid degradation in inflammatory or tumor environment, and significantly faster degradation rate when H2O2 is present.
The nano-grid BOF (N-BOF) is interwoven with tens of nanometer diameter fibers and has an open space of 10-300 nm, which has better liposome capture effect than spherical BOF (S-BOF) and can significantly change the biological distribution of liposomes in mice.The results show that free liposomes have little distribution in the lungs, fluorescence signal in the lungs after loading N-BOF is significantly enhanced, and lung bioavailability is increased by about 90 times.
In addition, N-BOF has a good capture effect on milk exosomes.Using TEM, the researchers observed that the exosomes loaded with N-BOF responsive release were 14% at pH 7.7 and 21% at pH 6.0 for 14 days, and that the exosomes remained intact.The results showed that N-BOF bungee could deliver liposomes to the lungs and prolong their time of action, and achieve exosome delivery and long-term release.
