L-tryptophan is one of the essential amino acids in the human body and is also a restrictive amino acid in animal nutrition.However, it is easily decomposed in acidic environments, causing great inconvenience to the quantitative determination of L-tryptophan in feed.The development of a biosensor for rapid detection of L-tryptophan in feed or animal body fluids will provide convenient means for the determination of L-tryptophan in animal nutrition.
In order to solve the above problem, the colloidal gold solution of surface modified peptides was synthesized by sodium borohydride reduction method, and the synthesis of nanomaterials and their sensitivity to L-tryptophan were verified spectroscopically.
The results showed that when L-tryptophan was added proportionally to the colloidal gold solution, the color of the solution changed from wine red to dark blue after 10 seconds of vibration mixing, and the maximum absorption wavelength of the solution moved from 535 nanometers to 600 to 620 nanometers.After optimizing the process conditions, the biosensor showed a good linear relationship to the gradient concentration of L-tryptophan between 1 and 50 microns and 50 to 1000 microns.
A colorimetric L-tryptophan biosensor based on peptides and gold nanoparticles was designed. It has the characteristics of simple structure and operation, high sensitivity and good selectivity.
The results of the study were published online in the Microchemical Journal on January 4.The research has been supported by the national key research and development plan, the National Natural Science Fund project, the China Postdoctoral Science Fund, and the construction of modern agricultural industry technology system.
