Zhang Peng, a professor at Tongji University School of Medicine / Affiliated Shanghai Lung Hospital, Zhou Hu, a researcher at Shanghai Institute of Pharmaceutical Research, and Ji Hongbin, a researcher at the Center of Excellence in Molecular Cell Science and Gao Daming.The findings were published online Jan. 5 in Cell, which is also the opening work of Chinese scholars in Cell 2024. The research team analyzed the tumor tissue and paired tumor tissue samples of 112 patients with small cell lung cancer, and systematically revealed the molecular features of small cell lung cancer by integrating genome, transcriptome, proteome and phosphorylated proteome. Further monitoring the proteomic data to screen prognostic biomarkers, the researchers found that the high expression of HMGB3 was closely associated with poor prognosis in patients and was validated by immunohistochemistry in independent cohort clinical samples.Mechanism studies have shown that HMGB3 can promote cell migration in small cell lung cancer through transcriptional regulation of cell-junction-associated gene expression. Secondly, the researchers systematically analyzed the immune microenvironmental characteristics of small cell lung cancer and found that the ZFHX3 mutation was closely related to the increase in immune cell infiltration levels.What's more, tumor samples from small cell lung cancer patients receiving PD-1 or PD-L1 inhibitors combined with chemotherapy have demonstrated a better therapeutic response in patients with ZFHX3 mutations, suggesting that ZFHX3 may be a biomarker for immunotherapy. Finally, the researchers divided small cell lung cancer into four subtypes using multi-histological data, systematically characterizing the unique molecular characteristics of each subtype and proposing potential treatment strategies.These molecular typing-guided specific therapeutic strategies were validated using mouse transplant tumor models derived from patient samples and mouse transplant tumor models derived from human small cell lung cancer cell lines. The research results provide theoretical basis for pathological mechanism analysis, prognosis detection, molecular typing and personalized treatment of small cell lung cancer.
