In 2006, American scientists Andrew Fire and Craig Mello won the Nobel Prize in Physiology or Medicine for their work on RNA interference.Today, the technology is increasingly used to regulate the expression of human genes. In mammalian cells, SIDT2 is involved in intracellular DNA/RNA autophagy and transports lysosomal nucleic acid to cytoplasm, triggering innate immune response, and SIDT1 directly mediates miRNA absorption.However, the molecular mechanisms of SID-1 transmembrane family proteins SIDT1 and SIDT2 mediated the absorption of heterologous small RNAs are still unclear, especially at low pH. The research team first solved the difficulties of membrane protein expression purification and the serious orientation advantage in the preparation of frozen samples.Through structural analysis, the researchers found that SIDT1 and SIDT2 were highly similar in structure, suggesting that they may have similar biological functions.In addition, the researchers further explored the molecular mechanisms of SIDT1 and SIDT2 mediated small RNA transport. The structure of transmembrane family proteins SIDT1 and SIDT2 in mammalian cells was analyzed. The molecular characteristics of pH-dependent binding and oligomerization of SIDT1 and SIDT2 with RNA were first proposed.Therefore, this study provides the molecular basis for SID-1 transmembrane family protein in nucleic acid transport, and also provides the theoretical basis for gene transboundary regulation.
