Up to 20 percent of adults worldwide suffer from migraines, which are recurrent and difficult to treat.Some people experience sensory symptoms before a headache attack, such as tingling in the body, while others do not.It is not clear why two types of migraines exist, either with or without signs.
Debbie Hay of the University of Otago, New Zealand, said: "It is well known that migraines are familial, but it is not easy to determine the genetic basis for each subtype.”
Now, Kári Stefánsson of deCODE genetics, an Icelandic biopharmaceutical company, and colleagues have identified genetic variants that affect whether people develop migraine headaches.
Researchers analyzed the DNA of 1.3 million people from Iceland, Denmark, the United Kingdom, the United States and Norway, of whom about 80,000 had migraines.
They found 44 genetic variants associated with the disease, 12 of which had never been described before.The team found that a rare variant of the PRRT2 gene, which helps regulate signal transmission between neurons, is associated with a high risk of premonitory migraine and epilepsy.
On the other hand, the rare variation in the function of SCN11A and KCNK5 genes appears to prevent both types of migraine, which play a role in the intercellular transport of sodium and potassium.
These findings may lead to new treatments for migraine causes, such as drugs that inhibit SCN11A and KCNK5 genes encoding proteins.
"This finding should give great hope to migraine sufferers."The current treatment does not completely eliminate the trend of migraine, so there is plenty of room for better treatment," Stefánsson said.”
