Recently, Shao Changwei, a researcher at the National Key Laboratory of Aquaculture Biological Breeding and Sustainable Production at the Yellow Sea Institute of Fisheries Research, China Academy of Fisheries Sciences.This is the first development and application of engineered exosomes in aquatic animals, which will play an important role in the precise regulation of gene function and reproductive stem cell transplantation.The research results were published in the journal Controlled Release in the field of drug delivery. Exosomes are extracellular vesicles that can be secreted by most cells. The diameter is about 100 nanometers. Because of their advantages of low immunogenicity and low toxicity, exosomes have become very promising functional molecules and drug delivery carriers.Shao Changwei's team successfully constructed an engineered exosome by bioinformatics, molecular and cell biological experiments, using the conserved sex-critical gene dMRt1 as the regulatory object. The engineered exosomes construction technology system mainly includes three parts: screening of intranets, determination of target peptides and preparation of engineered exosomes. The inclusion screening refers to the screening of miRNAs that can effectively inhibit the expression of dMRT1 sex-determining genes through experimentally.In order to determine the target peptide, we first select the receptor with high expression in the spermatid cell membrane by means of single cell sequencing and full transcriptome sequencing.The exosomes of ovary cells were extracted by differential hypercentrifugation, and the candidate peptides were loaded onto the surface of exosomes by phospholipid membrane anchoring technique.Finally, the expression of dMRT1 in spermatogenesis was inhibited by using the engineered exosome in vivo, which proved that dMRT1 plays an important role in maintaining spermatogenesis. The establishment of the technology system not only controls the function of sex-related genes, but also applies to the target transplantation of fish germ stem cells. The study is another important landmark in the field of epigenetics of fish sex determination and differentiation following the team's first confirmation of the regulatory mechanism of cRNA and the cytological basis of fish sex generation. The research was supported by the National Key R&D Plan, the National Natural Science Foundation Youth Science Foundation, the Taishan Scholars' Young Experts, the National High-level Talents' Special Support Plan.
