Autism (ASD) and schizophrenia (SCZ) are common chronic mental disorders with overlapping characteristics in behavior, genetics and neuropathology, suggesting that autism and schizophrenia may have common neuropathogenesis. Ageing is closely related to the dynamics of epigenetic effects, and changes in DNA methylation patterns have been observed in many age-related diseases.The tissue-specific "epigenetic clock" constructed on this basis can accurately predict the physiological age of samples, so it is called "epigenetic age".The deviation of epigenetic age from actual age can be used as an indicator to measure the rate of biological aging. Recent studies have found that there are systematic and age-related biological changes in the brains of patients with mental disorders such as schizophrenia.In response to the above problems, researchers at Xi'an Jiaotong University calculated the epigenetic age of the cerebellum, prefrontal cortex, temporal cortex, and cerebellum, striatum and hippocampus in 3 brain regions in patients with schizophrenia.The Horvath epigenetic clock consists of 353 CpG sites associated with aging. The DNA methylation data obtained from different platforms are standardized using the β mixed fraction standardization (BMIQ).Subsequently, the researchers established regression models of methylation age and actual age in each brain region to estimate the epigenetic age acceleration in each brain region.Epigenetic age is significantly correlated with actual age in different brain regions of patients with autism and schizophrenia, suggesting that epigenetic age can be used as an indicator of aging rate.The researchers further analysed the total sample and found that autistic patients over 45 showed an accelerated epigenetic senescence in cerebellar tissue and an accelerated epigenetic senescence in schizophrenic patients over 50.After sex stratification, no significant difference in epigenetic age acceleration between male and female patients was observed in different brain regions.These results suggest that the cerebellum region of patients with autism and schizophrenia deviates from the normal aging path, which provides a new scientific clue to the mechanism of autism and schizophrenia. The results were published online in the journal Molecular Psychiatry, an international leading journal on mental illness, under the title of Epigenetic Analysis of Cerebellar Abnormal Aging in the Elderly with Autistic Pedigree Disorder and schizophrenia.Liu Li, a Ph.D. student at School of Public Health, Xi'an Jiaotong University, is the first author.
