A newly published cancer research paper in the international famous academic journal Nature shows that the combination of immunotherapy with chemotherapy and targeted drugs (triple therapy) is expected to improve the outcome of patients with human epidermal growth factor receptor-2 protein (HER2) positive gastric tumors. The study, based on an interim analysis of phase III clinical trials, has prompted the U.S. Food and Drug Administration (FDA) to approve the clinical use of the therapy.
According to the paper, about 20% of patients with advanced gastric adenocarcinoma or gastroesophageal junction adenocarcinoma will express human epidermal growth factor receptor-2 protein. For more than 10 years, the standard first-line therapy has been the combination of anti-HER2 antibody trastuzumab and chemotherapy.
The corresponding author of the paper, Yelena janjigian of memorial Sloan Kettering cancer center and will Cornell Medical School in the United States, and colleagues evaluated the therapeutic benefits of adding pembrolizumab as part of the phase III clinical trial of keynote-811.
Pabolizumab is an antibody that can help the immune system recognize and attack tumor cells. The trial counted the efficacy data of 264 enrolled patients. The author's research team found that the addition of pabolizumab on the basis of trastuzumab and chemotherapy can reduce the tumor of some patients and induce complete response (temporary complete remission), the complete response rate can be increased from 3.1% to 11.3%, and the proportion of patients with the best response has also been significantly improved.
In view of the good results, the authors suggest that the same triple therapy should also be tested in early patients with the disease. In the future, the trial will continue to monitor the impact of this combination therapy on patient survival.
