After fertilization, the persistent calyx of Physalis in Solanaceae expands rapidly with the development of fruit, which forms the innovative structure of "Chinese lantern". Previously, he Chaoying's team isolated a Physalis pubescens "double-layer lantern" mutant doll1, which has petal sepal (with double-layer calyx) and stamen carpel. This is a typical B-function gene mutation. Research has confirmed that doll1 is indeed caused by the deletion of class B MADS box gene pfglo1 (doll1), and it is difficult to bear seeds after polling doll1 with wild-type pollen, This indicates that carpel function is significantly affected, but how class B MADS box gene regulates carpel development and its regulatory mechanism are still unclear.
After analyzing the mechanism of doll1 affecting carpel development, researchers found that doll1 affected carpel development and function at multiple levels, including morphological structure, pollen tube guidance function, ovule number and embryo sac development; This study further elucidated that doll1 ectopic expression in the carpel, and recruitment of carpel specific yabby transcription factor gene pfcrc as downstream gene is an important way for doll1 to achieve its new function in the regulation of carpel development.
The researchers also found that pfcrc is also a pleiotropic gene. In particular, the pfcrc gene editing mutant pfcrc obtained by crispr-cas9 gene editing technology specifically changed the development of carpel, that is, the ovary with 2 carpels healed turned into a fully open dorsal ventral lamellar structure, and the number became multiple (11-14), in a spiral arrangement, with 0-3 naked ovules at the base edge of each lamellar organ.
These results provide new evidence for understanding the functional evolution of class B MADS box genes and pfcrc genes in flower development, help to understand the formation of gene pleiotropy, and provide new ideas for understanding the origin and development of carpels.
